Bacterium transforms into weapon against sleeping sickness

ScienceDaily (Aug. 30, 2012) ? Scientists of the Antwerp Institute of Tropical Medicine (ITG) opened a new front against the cause of sleeping sickness. This parasite is transmitted between humans by tsetse flies. The researchers learned a bacterium living in those flies how to produce antibodies against the parasite. Application in the field is still a long way of, but the technique shows quite some promise.

Sleeping sickness is caused by trypanosomes, parasites being transmitted by the bite of a tsetse fly.

The World Health Organization estimates the yearly death toll at between 10,000 and 20,000 people. On top of that, the parasite also infects cattle, causing considerable economic loss. Many small African farmers depend on their cattle.

Without treatment, an infection is irrevocably fatal. Unfortunately, many poor people present at the hospital only in a late stadium. At that time the Trypanosoma parasites have lodged themselves in the brain, behind the notorious blood-brain barrier that keeps most drugs out. Arsenic compounds can pass the barrier and kill the parasite, but they also kill five per cent of the patients. New drugs are not in the pipeline.

Besides the parasite, one may also attack its vector, the tsetse fly. But insecticides may be detrimental to the environment, certainly in the long run. Therefore scientists look for alternative strategies. For instance genetically modified insects that are incapable of being infected by the parasite, or do not transmit it. But germline transformation of tsetse flies is unfeasible. To do so, one must be able to handle the eggs, but tsetse flies do not lay eggs, they directly bring forth a larva.

Therefore, the Antwerp researchers took another road. Tsetse flies harbour, as is the case with many insects, resident bacteria. One of them, Sodalis glossinidius (literally: companion of the tsetse fly) exclusively lives in tsetse flies. And it can be cultivated in the lab. De Vooght was the first to genetically modify the bacterium so it produces, and excretes, a very efficient type of antibody, called a nanobody. She identified two different secretory pathways that transported the nanobodies out of the bacterium. She also demonstrated that the bacterium was not hampered by its modification, so it can stand its ground amidst non-modified, 'wild type' congeners inside the fly.

Next, with antibiotics she cleared tsetse flies of their wild type bacteria and replaced those by the modified bacteria. These successfully colonized the flies and started producing nanobodies. The nanobodies also were present in the midgut, where the sleeping sickness parasite also is to be found.

De Vooght demonstrated the feasibility of the technique, but it still needs some development before it can be used to control sleeping sickness in the field. For instance, the antibodies now produced by the bacteria, are directed against a form of the parasite occurring in humans, not in flies. This is simply because this antibody was available, while the one against the fly form still has to be developed. De Vooght: "We wanted to demonstrate first that the technique works in principle. Now we have achieved that, we can tackle the technical details."

To the scientists it is just as important that symbiotic bacteria producing all kinds of substances are a means to getting insight into the interactions between disease-causing organisms and their insect vectors. The Antwerp researchers already demonstrated that the sleeping sickness parasite interferes with the saliva production of tsetse flies, forcing them to bite more humans than they otherwise would do. Insight in that kind of interactions might be instrumental to opening new ways of attacking diseases.

Bellevue College wins $550k NSF grant to improve STEM education

Bellevue College has been awarded a $550,000 grant from the National Science Foundation (NSF) to fund the next phase of a project that is changing the way biology is taught at community colleges throughout the state. By immersing students in the actual practice of scientific research, the project expects to boost the number of students who pursue STEM (science, technology, engineering and math) majors and careers.

The effort is based on a program created at BC several years ago, called ComGen: Community College Genomics Research Initiative, one of the pioneering community college research projects in the nation. Students perform original research by sequencing the genome of a bacterium that fights a wheat fungus. They also analyze primary research articles and interact frequently with scientists.

These research opportunities for undergraduates are common at research universities, but much less so at community colleges, where up to half of all students are introduced to STEM fields.

?Our students at BC have really benefited from the opportunity to perform real research and then share it with the wider scientific community. They gain a much better understanding of biology compared to just doing recycled lab exercises,? said Principal Investigator and Assistant Dean of Sciences Gita Bangera, Ph.D., who created the ComGen project. She has conducted research in bacterial molecular biology at Harvard University, UW, Washington State University and the University of Copenhagen (Denmark).

BC has received over $11 million in NSF grants since 1995. With BC?s lead, Tacoma Community College, Everett Community College, Clark College (Vancouver) and Olympic College (Bremerton) will initially implement the program, with plans to recruit more of Washington state?s 34 community and technical colleges.

?Ultimately, this project will open the door to opportunities in the sciences for many more nontraditional students, who make up a large part of student bodies at community colleges, such as people of color, older students, parents, veterans and those with low-income backgrounds,? said Rob Viens, Ph.D., dean of BC?s Science Division.

Chronic stress linked to high risk of stroke

ScienceDaily (Aug. 29, 2012) ? Chronic stress, prompted by major life stressors and type A personality traits, is linked to a high risk of stroke, finds research published online in the Journal of Neurology Neurosurgery and Psychiatry.

Chronic stress, manifested as physical and/or mental symptoms in response to stressors lasting longer than 6 months has been linked to a heightened risk of heart disease. But its impact on the risk of stroke has not been clear.

The research team base their findings on150 adults, with an average age of 54, who had been admitted to one stroke unit, and 300 randomly selected healthy people of a similar age who lived in the same neighbourhood.

Levels of chronic stress were assessed using the combined quantitative scores of four validated scales, looking at major life events; symptoms, such as anxiety and depression; general wellbeing; and behaviour patterns indicative of type A personality (ERCTA scale).

Type A behaviours include hostility, aggression, impatience and a quick temper. A score of 24 or higher on the ERCTA scale is considered to be indicative of a type A personality.

Participants were also assessed for known biological risk factors for stroke, including diabetes, high blood pressure, high cholesterol, any history of heart rhythm abnormalities, and daytime sleepiness.

And they were quizzed on their lifestyle, including their caffeine, alcohol and energy drink intake, as well as whether they smoked, had a partner, and a job.

The results showed that several factors were independently associated with a heightened risk of stroke.

Compared with the healthy comparison group, the risk of a stroke was almost four times higher among those who had experienced a major life event in the previous year.

A high score on the ERCTA scale more than doubled stroke risk, as did a current or previous history of smoking, and intake of two or more energy drinks a day.

And those with heart rhythm disturbances were more than three times as likely to have a stroke while those with a high daytime sleepiness score almost tripled their risk. And being a man boosted the risk nine-fold.

But when all the factors were assessed together, the risk of a stroke was associated with a stressful life and type A behaviours. And this held true, irrespective of other risk factors, including gender and an unhealthy lifestyle.

    'Nano machine shop' shapes nanowires, ultrathin films

    ScienceDaily (Aug. 29, 2012) ? A new "nano machine shop" that shapes nanowires and ultrathin films could represent a future manufacturing method for tiny structures with potentially revolutionary properties.

    The structures might be tuned for applications ranging from high-speed electronics to solar cells and also may have greater strength and unusual traits such as ultrahigh magnetism and "plasmonic resonance," which could lead to improved optics, computers and electronics.

    The researchers used their technique to stamp nano- and microgears; form tiny circular shapes out of a material called graphene, an ultrathin sheet of carbon that holds promise for advanced technologies; and change the shape of silver nanowires, said Gary Cheng, an associate professor of industrial engineering at Purdue University.

    "We do this shaping at room temperature and atmospheric pressure, like a nano-machine shop," said Cheng, who is working with doctoral students Ji Li, Yiliang Liao, Ting-Fung Chung and Sergey Suslov and physics professor Yong P. Chen.

    Graphene and nanowires -- filaments 1,000 times thinner than a human hair -- have numerous potential applications. However, technologies are needed to tailor them for specific uses. The new method, called laser shock-induced shaping, makes it possible to tune nanowires by altering electrical and optoelectrical properties that are critical for electronic components.

    The researchers also have shown how laser shock-induced shaping can be used to change the properties of graphene, a step toward harnessing the material for electronic applications.

    Findings were detailed in research papers published in the journal Nano Letters, and the work also was highlighted earlier this month in the News and Views section of the journal Nature Photonics.

    The technique works by using a multilayered sandwich structure that has a tiny mold at the bottom. Nanowires were situated directly above the mold, and other materials were layered between the nanowires and a glass cover sheet. Exposing this layered "forming unit" to an ultra-fast pulsing laser causes one of the layers to burn up, generating a downward pressure that forces the nanowires into the mold and changes their shape.

    "The process could be scaled up for an industrial roll-to-roll manufacturing process by changing laser beam size and scanning speed," Cheng said. "The laser shock-induced shaping approach is fast and low-cost."

    Part of the research, funded by the National Science Foundation, was carried out in a specialized clean room at the Birck Nanotechnology Center in Purdue's Discovery Park.

    Most of us who scrapbook are storytellers ? we want to tell the stories that matter, the stories that mean something to us. And I cannot think of a better way to tell the stories of your family than to create heritage scrapbook pages. Heritage scrapbook pages allow you to capture the stories of your family and helps to connect you to the past. I love researching my family history and then creating pages to document the past. I love learning how different their lives were from mine today as well as how the things that they experienced have affected our family traditions. It meets my own need to build meaningful connections between the past, today and our futures. I thought that we?d take a look at some basic tips for creating amazing heritage pages and then look at a bunch of heritage pages to help get you inspired to document your OWN family history!

    First, let?s talk about some basics -

    1. Organization of photos and stories from the past is critical when you decide you want to begin documenting your family?s history. Being organized up front will make the creation of your pages so much easier later! I would suggest that you gather your photos, stories and ephemera (the ?stuff?) and organize them in a way that makes sense to you. Personally, I?ve chosen to organize my own heritage stuff by person ? I have a folder for each of my ancestors and put all of the relevant photos, ephemera and stories into that folder. Then, when I?m ready to create a page about a particular person or event, I know right where to go to get all of the details!

    2. Regardless of how you intend to scrapbook your family history ? digitally or with paper ? I strongly recommend that you have ALL of your priceless family photos scanned into digital images. Then, you can be assured that nothing you do while creating heritage pages will hurt your photos in any way. This will also allow you to share these photographs with other family members who may be interested in having a copy!

    3. Don?t be afraid to scrapbook the family photos or stories! Yes, they are precious ? but don?t let that stop you! In fact, let that be the very reason for getting them documented as soon as possible! They are precious and deserve to be told!? Nothing you create will do anything but give them the attention they deserve!? Push past any reservations you might have and JUST DO IT!

    And now for a few general tips for scrapbooking heritages pages -

    1. Tell those stories and be sure to include any details you know about what was happening! Journaling is key for heritage pages ? even if its just a few details! Also, don?t let not having a photo keep you from telling the story ? create a photo-less page or find elements that help support the story you want to tell. Use the historical or cultural details of the time to help tell your stories as well! Let me give you an example ? we know that my grandfather was present during the bombing of Pearl Harbor but I don?t know any details beyond his being present & the impact his presence at that historical even had later on in his own family. But, I did some research and created a page all about the bombing of Pearl Harbor since we know he was there for such a major historical event! That is an important story about our family even if I don?t have photos or details!

    Venezuela firefighters struggle with refinery blaze

    PARAGUANA, Venezuela (Reuters) - Venezuelan firefighters struggled to finish putting out a blaze at the country's biggest refinery on Tuesday, with a fuel tank twice bursting into flames within minutes of authorities declaring it fully extinguished.

    The partly-melted storage tank burned on and off at the 645,000-barrel-per-day Amuay refinery as teams poured foam over it to suffocate a fire sparked by a pre-dawn blast on Saturday that killed 48 people.

    The blaze had been put out at around 10:30 a.m. (1500 GMT). Though it was not immediately clear if it might break out again, the fire seemed to be largely under control.

    Energy Minister Rafael Ramirez has said the facility should restart within two days of the fire being put out. None of the processing units were hit by the blaze, though refinery operations were shut down on Saturday for safety reasons.

    Ramirez has repeatedly assured that Venezuela has sufficient fuel inventories to supply the domestic market and continue meeting export commitments.

    U.S. gasoline futures dropped sharply on Tuesday, a day after rising as much as 3 percent on the combination of the Amuay fire and Tropical Storm Isaac's potential threat to U.S. to Gulf Coast refinery operations.

    The blast flattened hundreds of homes and left dozens wounded. Many of the dead were National Guard troops who were providing security in a compound next to the tanks.

    The charred remains of two other fuel tanks, which had been extinguished before dawn, were almost entirely melted from three days of the blazing inferno.

    Traders say the impact on fuel markets may continue even after Amuay is up and running again. Tank farm accidents often cause problems with gasoline blending, which may leave PDVSA relying on imports.

    Venezuela has for years had to import fuel due to recurrent refineries outages.

    Chavez said at the scene on Monday that he was creating a fund worth about $23 million to help pay for clean-up operations and replace homes destroyed by the pre-dawn blast.

    It was one of the most deadly oil industry accidents in recent years, nearing the toll of the 1997 fire at India's Visakhapatnam refinery that killed 56 and topping the 2005 blast at BP Plc's Texas City refinery in which 15 people died.

    Defense Logistics Agency to purchase six ... - Food and Drink Digital

    PHILADELPHIA, Aug. 27, 2012 /PRNewswire-USNewswire/ -- At the direction of the president, the Defense Logistics Agency is procuring a six-month advanced meat supply for its Department of Defense customers, to provide relief to the drought-stricken agriculture industry.

    The DLA Troop Support Subsistence supply chain is asking its top three prime vendors to purchase, store and distribute beef, pork, lamb, chicken and catfish items, said Tom Daley, deputy director. He estimates purchases will amount to more than $100 million.

    In 2011, Subsistence provided 95 million pounds of beef, 68 million pounds of pork, 500,000 pounds of lamb, 55 million pounds of chicken and 2 million pounds of catfish to the U.S. military services and other DOD agencies all over the world.

    "Dining facility menus will not be affected by this purchase," Daley said. "A soldier at a DFAC in Afghanistan won't notice anything different when eating a grilled cheeseburger or baked chicken. The warfighter will continue to receive healthy, wholesome and nutritious meals."

    Subsistence officials are negotiating with prime vendors on additional storage and distribution fees, estimated to be between $2.4 and $3.3 million. As they do for all food products, the prime vendors will store the additional meat, which will be frozen to maximize shelf-life.

    Purchases will be based on forecasted demand. Subsistence is also working with the suppliers to identify the specific meat items to purchase.

    "We are focusing on high-demand items with long shelf-life and items that will make an immediate impact on the industry," Daley said.

    Subsistence is working with the Joint Subsistence Policy Board to keep the military services informed as it finalizes purchase details. Officials are also working with the board, made up of service food advisors, to determine standardized meat pack sizes and other details.

    KAMPALA, Uganda (AP) ? Uganda's president has made his son a one-star general and appointed him commander of the country's special forces, leading some Ugandans to conclude that the son is being groomed to succeed his father.

    New Novartis drug shows potential in heart failure treatment

    MUNICH (Reuters) - An experimental drug from Novartis may help up to half of heart failure patients for whom no effective treatment is available, although the evidence so far is indirect.

    Results of a mid-stage clinical trial of the drug known as LCZ696, unveiled on Sunday, gauged its effectiveness by measuring whether patients had lower levels of a protein linked to the debilitating condition.

    Researchers said that the drug successfully reduced levels of the protein NT-proBNP, although they cautioned that larger studies were needed to see if this "biomarker" signal would translate into better outcomes in clinical practice.

    Novartis, long a major player in cardiovascular medicine, is looking to LCZ696 as one of two new treatments for heart failure that could revive its fortunes at a time when top-selling blood pressure pill Diovan is facing generic competition.

    The Swiss company said it had not yet decided whether to advance LCZ696 into final-stage testing for the particular group of patients studied in the latest trial. However, Laurie Letvak of its critical care unit said the results were "a great first step".

    The U.S.-led study looked at a hard-to-treat group of patients with so-called preserved ejection fraction heart failure. These patients have heart chambers that are stiff but are still able to pump a reasonable amount of blood, as measured by ejection fraction.

    Heart failure is a condition where the heart struggles to pump blood around the body. Unlike a heart attack, in which a heart artery becomes blocked, it develops progressively.

    People with preserved ejection fraction are still able to pump more than 40 percent of blood from the heart's left ventricle, against 55-70 percent for a healthy heart.

    MORE WORK TO DO

    Patients given LCZ696 showed sharp reductions in NT-proBNP after 12 weeks compared to those on Diovan, although the difference was no longer significant by 36 weeks, Dr Scott Solomon of the Brigham and Women's Hospital told the annual meeting of the European Society of Cardiology.

    Encouragingly, the size of the heart was also reduced in patients on the new drug - a good sign in heart failure.

    Solomon, whose findings are being published online by The Lancet medical journal, was cautious about reading too much into the results, saying simply "further testing of this compound could be warranted".

    Other experts not involved in the research were also wary.

    "To date, there's been no intervention that has been helpful for this patient group, so this is at least a signal," said Dr Mariell Jessup, medical director of the Penn Medicine Heart and Vascular Center in Philadelphia.

    "It's interesting but there's lots more work to do."

    LCZ696 is a combination of Diovan, or valsartan, and another class of drug called a neprilysin inhibitor.

    Neprilysin inhibitors have had a chequered history, with Bristol-Myers Squibb suffering a major setback 10 years ago when its product Vanlev flopped due to cases of angioedema, a swelling of the face, neck, lips and throat.

    LCZ696 was well tolerated in the latest Novartis-funded study, which involved 301 patients.

    Novartis is also testing the medicine in another group of heart failure patients with reduced ejection fraction, as well as assessing its role in fighting high blood pressure.

    The company has a second experimental drug for acute heart failure called RLX030 in final Phase III trials, for which clinical trial results are expected later this year.

Wii players can smack a tennis ball by swinging their hands when they hold a wireless remote. They can jog in place, climb a virtual mountain, or shoot menacing robots in the same way. This is known as gesture sensing. The tech isn?t quite ready for standard computing yet. However, it?s not difficult to imagine a future in which gesture sensing becomes one of the primary modes of interacting with our computers.

    Multi-touch technology

    But this is just one interaction innovation. There?s also multi-touch technology. Again, iPad and other tablet users are already familiar with this. By tapping icons on their screen, they can open apps and programs. By swiping their fingers across the screen, they can flip to a new page on a website. By pinching pages, they can zoom on an image to make it larger.

    Voice recognition

    Another likely tech jump involves voice recognition. Rather than clicking a mouse button to open a program, users can simply tell their computers to open a particular word file or close iTunes. Some of this already exists, most notably Siri, the personal assistant built into the iPhone 4S.

    No matter what happens, though, the odds are that we?ll still rely at least somewhat on the humble mouse. After all, it?s served us well for many years.

    GamrCred: A New Type of Gaming Community | Gaming Capacity

    GamrCred is a place solely built for a gaming community. We were invited to the beta and came away a little impressed with what they are trying to achieve here. First let?s talk about what they hope to achieve.

    GamrCred is all about what you play and say to earn respect from other people. You can connect your gaming accounts and your social accounts to GamrCred and people can click the respect button on stuff you say and what trophies you earn if the deem fit. Play + Say + Respect = GamrCred.

    On sign up, after the usual stuff, I?m invited to add any gaming profiles. The gaming profiles available at the moment are PSN, Xbox Live and Steam accounts. All the games and achievements are then added to your GamrCred account for people to see and also boost your Play rating. The more games you pay and the more acheivements/trophies you earn the better the Play rating. Then I?m asked if I want to add social profiles like Facebook and Twitter to be connected to you GamrCred account. The whole process was simple and quick and will not deter the people even with the smallest amount of patience.

    Once in, you can invite your friends and make new friends. Compare you trophy collection with others and even give them, like mentioned earlier, a ?Respect? for trophies that they?ve earned. What you say on you social accounts can also gain respect so it?ll encourage you to be more vocal about your games. You can see exactly how well you are doing in each category ? Play, Say, Respect ? from slider bars on the left of you profile for each category. Depending how well you are doing, the sliders will move to give you a better or worse rating which in turn then gives you a GamrCred rank depending on your performance. To bolster up the community just a little bit for you have the standard forum where you can do the usual forum things.

    The experience was a positive one and the idea of a massive gaming community linking all their gaming profiles to show off their achievements is kind of is exciting. As you would expect in beta mode there is a few bugs and maybe a few areas that they can improve, like navigating seemed a bit of a chore sometimes. That said, they have got the basics right and GamrCred has great potential.

    Quake catalog details Europe's future risks

    Earthquakes in Europe have influenced everything from its legends to its languages.

    According to Greek mythology, the Oracle at Delphi spoke through women who inhaled delirium-causing vapors; now, modern geologists say these vapors were hydrocarbon gases released along earthquake faults and fractures crisscrossing the site of the Delphi temple.

    In Italy, a magnitude-6.7 quake in 1638 permanently tweaked the Calabria region's dialects, according to a study published in the journal Annali di Geofisica in 1995. For example, the town of Savelli, founded after the quake, was linguistically isolated from its neighbors because it was settled by refugees from villages far to the west.

    About 45,000 earthquakes large enough to feel have rocked the continent in the past 1,000 years, according to a newly updated catalog of earthquakes in Europe and the Mediterranean. Combining this historic information with modern-day geologic investigations is the first step in forecasting Europe's future risk of earthquakes, researchers say.     "One has to bring together the information from very old times, and from very modern times," said Gottfried Gr?nthal, of the GFZ German Research Center for Geosciences in Potsdam, Germany.

    Estimating earthquake risk requires "good knowledge about the seismological past. That means we have to extend our knowledge of the seismicity of an area as long into the past as possible," Gr?nthal said.

    The new catalog reveals that most of Europe's seismicity is concentrated along plate boundaries in the Mediterranean, including the present-day countries of Italy, Greece and Turkey, along with regions in southern Iberia, the Balkans and the Caucasus.

    But earthquake risk is also found in more northerly locations, such as along the stupendous mountain-building faults of the Alps. One of Europe's largest and most devastating quakes of the past millennium was actually in Switzerland. The Basel earthquake, which struck on Oct. 18, 1356, with a magnitude 6.6, destroyed the town and toppled buildings in Germany and France.

    Scientists pinpointed the fault responsible in 2001, and concluded the next temblor there could occur in about 1,500 to 2,500 years.

    Building the catalog required Gr?nthal and colleague Rutger Wahlstr?m to act as both historians and seismologists. In central Europe, they collaborated with historians at the University of Potsdam to estimate the size of past earthquakes in Germany. For other regions, Gr?nthal relied on countries' own earthquake catalogs ? 80 in total. The researchers also examined more than 100 studies and performed their own analyses of key historical earthquakes.

    The bulk of the work involved standardizing the earthquake records to the modern "moment magnitude" scale, to ensure that a quake in 17th-century Germany could be compared to a quake in 16th-century Italy. The moment magnitude scale provides estimates of the total energy released during earthquakes, and replaced the Richter scale in the early 1980s.

    Magnitude is also a key part of the equations that predict ground motion, or shaking, which engineers and architects rely on to design earthquake-safe bridges and buildings.

    With each country's records brought up-to-date, researchers around the world can now use the catalog when assessing Europe's risk of future quakes. "Having a standardized earthquake catalog is the first step in developing a seismic hazard assessment," Gr?nthal said.

    A report describing the work appears in the July issue of the Journal of Seismology.
